You can right click on a layout tab to access the Page Setup Manager or the Plot dialogue box.  The easiest thing to do for page setups is skip the extra dialogue box for the Page Setup Manager and make your page setups directly in the Plot dialogue box.  Open the Plot dialogue, make your setups, and then pick the Apply to Layout button to save the setup.  If you don't want to print, simply pick the Cancel button to save the setup and not print to the specified device.
